Preparing a Semen Sample

Before running a semen analysis with your Ongo Compact, it's essential to properly prepare the semen sample. This article will guide you through the process of preparing a semen sample for analysis, including recommended dilution rates for various species and tips for handling and homogenizing the sample.

Dilution Rates for Optimal Analysis

Ongo Compact is designed to analyze semen samples within a specific concentration range for accurate results. The optimal range is 10-100 million sperm cells/ml, with an extended range of 5-120 million sperm cells/ml.

To achieve the desired concentration, dilute the raw semen according to the following table:

Sample Handling and Homogenization

To ensure accurate and consistent results, follow these tips for handling and homogenizing your semen samples:

  1. Gentle handling: Handle semen samples gently to avoid damaging the sperm cells. Avoid exposing the sample to extreme temperatures or excessive agitation.
  2. Homogenization: If using a large quantity of diluted sperm (e.g., 100 ml of pig sperm), homogenize the sample before testing. Gently mix the sample by inverting the container 3-4 times.
  3. Timely analysis: Analyze the semen sample promptly after collection and preparation to minimize the effects of time on sperm quality.
  4. Clean equipment: Use clean, sterile equipment for semen collection, dilution, and handling to prevent contamination and ensure accurate results.
  5. Consistent dilution: When diluting the semen sample, ensure that the diluent is well-mixed with the semen to create a homogeneous solution. Use a pipette or other precise measuring device to achieve consistent dilution rates.

Preparing the Sample for Analysis

Once you have diluted and homogenized your semen sample, you are ready to load it onto the Ongo Slide for analysis:

  1. Gently mix the diluted semen sample by inverting the container 3-4 times.
  2. Using a pipette, transfer 5 μl of the diluted semen sample to the right-hand chamber of the Ongo Slide.
  3. Ensure that the sample is placed above the arrow and into the U-shaped part of the slide.
