Running a Semen Analysis

With your semen sample prepared and diluted, you're now ready to run a semen analysis using your Ongo Compact. This article will provide step-by-step instructions on loading the test slide, starting the analysis, adjusting focus and lighting, selecting an optimal field of view, and repeating measurements for improved accuracy.

Loading the Test Slide

  1. Ensure your Ongo Compact is connected to a power source or sufficiently charged.
  2. Gently open the slide stage protection lid (maximum angle: ~95-100°).

    CAUTION: Do not push the lid beyond its maximum angle, as this may cause damage.

  3. Carefully lift the pivoting illumination unit until you hear a clicking sound (maximum angle: ~75-80°) CAUTION: Do not lift the illumination unit beyond its maximum angle, as this may cause damage.
  4. Place an Ongo Sperm Test slide on the slide stage and gently push it until it reaches the mold edge under the illumination unit. The right-side chamber of the slide should be above the camera lens hole.
  5. Carefully push the pivoting illumination unit back down over the positioned slide.

Starting the Analysis

  1. From the main screen, select "Start" (press the left end button). The slide stage will begin warming up.
  2. The current temperature of the heated slide stage will be displayed in the lower-left corner of the screen. The target temperature is preset based on the selected animal species.
  3. When the slide stage reaches the preset temperature, a slide checking procedure will begin automatically. If the slide is properly positioned, a live view of the sample will appear on the screen, and Ongo Compact will start autofocusing.

Adjusting Focus and Lighting

Ongo Compact's autofocus mechanism generally provides good results, but you can manually adjust the focus and lighting if needed:

  1. Focus: Use the "Focus +" (right middle) and "Focus -" (right end) buttons to adjust the focus. The current focus value is displayed in the top-right corner of the screen.
  2. Lighting: Use the toggle button (left middle) and the "Light +" (right middle) and "Light -" (right end) buttons to adjust the lighting for optimal contrast. The preset value is usually close to optimal.

Note: Sperm cells should appear as small, dense black spots against the background.

Selecting an Optimal Field of View

  1. Choose a field of view without debris or bubbles.
  2. Ensure the long side of the slide is pushed up against the housing for correct slide placement.
  3. Move the slide slightly up or down on the heated slide stage to view different areas of the sample

Analyzing and Repeating Measurements

  1. Press "Continue" to start the analysis. After 30-40 seconds, the results will be displayed.
  2. Select a new field of view by moving the slide slightly, then press "Repeat" (left middle button).
  3. Repeat the process at least 2 more times to improve statistical accuracy. Aim to capture around 1,000 cells in total, as indicated on the results screen.
  4. Press "Finish" to save the report and videos to the pen drive.

CAUTION: Always repeat the measurement on the same sample at least 3 times for accurate results.
