Proper semen sample preparation is crucial for obtaining accurate results with your Ongo Vision device. This article will guide you through the steps to correctly collect, store, and load a semen sample onto an Ongo Slide, as well as the necessary dilution procedures to ensure optimal analysis conditions.

Sample Collection

  1. Collect fresh, cooled, or post-thaw semen samples in a clean, sterile container.
  2. If using a collection condom, make sure it is specifically designed for semen collection and does not contain spermicidal lubricants.
  3. Collect the entire ejaculate, as the sperm concentration and motility may vary throughout the sample.
  4. Note the time of collection, as semen samples should be analyzed within 1 hour of collection for optimal results.

Sample Storage

  1. If the sample cannot be analyzed immediately, store it at room temperature (20-25°C or 68-77°F) for up to 1 hour.
  2. Avoid exposing the sample to extreme temperatures, direct sunlight, or excessive vibration.
  3. In case of fresh semen, do not refrigerate or freeze the semen sample, as this can damage the sperm cells.

Sample Dilution

  1. To adhere to the technological limitations of CASA for cell density assessment, ensure semen samples are diluted to fall within the range of 5-100 million sperm cells/ml, with an optimized range of 10-80 million sperm cells/ml.
  2. Utilize the suggested dilution rates based on species-specific semen density.
  3. For large volume samples, such as 100 ml pig sperm, homogenize the liquid by performing 3-4 slow upside-down movements to ensure even distribution.
  4. CAUTION: Accuracy has been tested only in transparent mediums. Using milk or other particle-dense diluents could compromise accuracy.

Slide Preparation

  1. Place an unused Ongo Slide on the heated stage's periphery to warm it to body temperature, ensuring the chambers are upside down.
  2. Use appropriate tools to deposit about 5 microliters of the diluted semen sample at the chamber's opening, indicated by an arrow. The fluid will automatically be drawn into the chamber.
  3. Remove any excess fluid with a cloth to prevent sample drift.

Tips for Optimal Results

  • Ensure that all collection and storage containers are clean and sterile.
  • Avoid using samples that have been contaminated with lubricants, soaps, or other substances.
  • In case of diluted fresh semen, you can calculate back the ejaculate concentration using the built-in calculator on the result page of the Ongo Vision App.
